ABSTRACT

Abstract MiRNA (or microRNA) is a subclass of non-coding RNAs that is responsible for post-transcriptional gene regulation. It has approximately 22 nucleotides and regulates gene expression in plants and animals at the post-transcriptional level, by the cleavage of a target mRNA or by suppression of its translation. Although many of the processes and mechanisms have not yet been fully elucidated, there is a strong association between miRNA expression and several diseases. It is known that miRNAs are expressed in the cardiovascular system, but their role in cardiovascular diseases (CVDs) has not been clearly established. In this non-systematic review of the literature, we first present the definition of miRNAs and their action at the cellular level. Afterward, we discuss the role of miRNAs as circulating biomarkers of CVDs, and then their role in cardiac remodeling and atherosclerosis. Despite the complexity and challenges, it is crucial to identify deregulated miRNAs in CVDs, as it allows a better understanding of underlying cellular and molecular mechanisms and helps in the development of more accurate diagnostic and prognostic circulating biomarkers, and new therapeutic strategies for different stages of CVDs.


Resumo O miRNA (ou microRNA) constitui uma subclasse de RNAs não codificantes responsáveis pela regulação gênica pós-transcricional. Ele possui aproximadamente 22 nucleotídeos e regula a expressão gênica em plantas e animais ao nível pós-transcricional, pela clivagem de um mRNA alvo ou da repressão de sua tradução. Embora muitos processos e mecanismos ainda não estejam completamente elucidados, existe uma forte associação entre a expressão de miRNAs e diversas doenças que acometem o organismo. Os miRNAs são expressos no sistema cardiovascular, contudo o seu papel no desenvolvimento das doenças cardiovasculares (DCVs) ainda não está totalmente elucidado. Diante disso, realizou-se uma revisão não sistemática da literatura a fim de se discutir a relação entre os miRNAs e as DCVs. Nesta revisão, primeiramente é discutido o que são os miRNAs e a sua ação a nível celular. Após, é discutido o papel dos miRNAs como biomarcadores circulantes de DCVs e então o seu papel no remodelamento cardíaco e na aterosclerose. Apesar da complexidade e dos desafios, a identificação dos miRNAs desregulados nas DCVs é crucial, uma vez que possibilita uma melhor compressão dos mecanismos celulares e moleculares envolvidos, assim como auxilia o desenvolvimento de marcadores circulantes de diagnóstico e prognóstico mais acurados e de novas estratégias terapêuticas para os diferentes estágios da DCV.

ABSTRACT

Handmade anatomy models may be a complementary useful tool to dissection and prosection, since the student interacts with the body structure in a three-dimensional way. In this study homemade dough (biscuit) was used to create a brain model. Two anatomy trainees from the medical school of the Universidade Federal Fluminense (Niteroi Brazil) were challenged to model the gross anatomy of the lateral aspect of the brain. They prepared and handled homemade dough to produce a simple and low cost model of the cerebral hemisphere. Styrofoam balls fixed by pins were used to create a framework, and dough rolls were modeled in order to create the brain sulci and gyri. At the end, the model closely resembled three-dimensional features of the human brain. Also, brain lobes were easily identified, as well as some major gyri and sulci, such as the central and lateral sulci and the precentral and postcentral gyri. In conclusion, the three-dimensional spatial characteristics, the clear identification of sulci and gyri and the brain lobes, make this model a good tool for students who only require basic anatomy in their curriculum.


Los modelos de anatomía elaborados a mano pueden ser una herramienta útil y complementaria a la disección y la prosección, debido a que el alumno interactúa con la estructura del cuerpo de manera tridimensional. En este estudio se utilizó una masa casera (bizcocho) para crear un modelo cerebral. Dos estudiantes de anatomía de la Facultad de Medicina de la Universidade Federal Fluminense (Niteroi, Brasil) fueron desafiados a modelar la anatomía macroscópica de la cara lateral del cerebro. Prepararon y manejaron masa casera para producir un modelo simple y de bajo costo del hemisferio cerebral. Bolas de espuma de poliestireno fijado por alfileres se utilizaron para crear un marco, y los rollos de masa fueron modelados con el fin de crear los surcos cerebrales y giros. Al final, el modelo resultó ser muy semejante a las características tridimensionales del cerebro humano. Además, los lóbulos cerebrales fueron fácilmente identificados, así como algunos giros y surcos mayores, tales como el surco central y lateral y el giro precentral y postcentral. En conclusión, las características espaciales tridimensionales, la identificación clara de los surcos y giros y los lóbulos cerebrales, hacen de este modelo una buena herramienta para los estudiantes que sólo requieren anatomía básica en su plan de estudios.

ABSTRACT

Quantitative morphology is a reliable tool in developmental, clinical and aging biology. Quantitative data of histological improvement and/or impairment due to dietary manipulations or pharmacological interventions draws the attention of countless researchers worldwide. Morphometry allows a wide range of two-dimension analysis to be performed, whereas image segmentation allows the measurement of a single structure or the determination of the intensity of its color after adequate immunostaining or even the area occupied by a specific sort of cell. When it comes to unbiased analysis, consideration should be given to suitable tissue perfusion, fixation and embedding, avoiding artifacts. Furthermore, tissue shrinkage should be taken into account. In order to ensure reproducibility, one of the most important principles in scientific research, and to allow different groups to be compared, image acquisition and image segmentation emerge as crucial steps and should be standardized. This paper aims to approach some important care for appropriate tissue processing and evaluation, and shows some practical examples of how morphometry and image segmentation can be applied to your experiment.


La morfología cuantitativa es una herramienta confiable en la biología del desarrollo, clínica y en el envejecimiento. Los innumerables datos cuantitativos de mejoras histológica y / o deterioro debido a las manipulaciones dietéticas o intervenciones farmacológicas atraen la atención de los investigadores en todo el mundo. La morfometría permite realizar una amplia gama de análisis en dos dimensiones, mientras que la segmentación de imágenes permite la medición de una única estructura o la determinación de la intensidad de su color después de una adecuada inmunotinción o incluso el área ocupada por un tipo específico de célula. Cuando se trata de un análisis imparcial, debe considerarse la posibilidad de la perfusión tisular adecuada, fijación e inclusión, evitando artefactos. Además, se debe considerar la contracción del tejido. Con el fin de asegurar la reproducibilidad, uno de los principios más importantes en la investigación científica, y para permitir que diferentes grupos se comparen, la adquisición y segmentación de la imagen aparecen como pasos cruciales y deben ser estandarizados. En este trabajo se abordan algunos cuidados importantes para el procesamiento de tejidos y evaluación apropiadas, y se muestran algunos ejemplos prácticos de cómo la segmentación morfométrica y la imagen se pueden aplicar a su experimento.

ABSTRACT

Se estudiaron con herramientas morfológicas, los efectos de diferentes dosis de Losartan sobre el remodelamiento cardiovascular, en ratas deficientes en óxido nítrico. 30 ratas Wistar, con 15 semanas de edad, fueron separadas en 6 grupos. control (C), L-NAME (LN), y 4 grupos en que administró LN junto con Losartan, en diferentes dosis (1, 5, 20 y 40 mg/kg/dia). El L-NAME fue administrado durante 9 semanas y la administración de Losartan se inició en la segunda semana de experimentación. Se estudiaron el corazón, la parte torácica de la aorta y la arteria mesentérica craneal, con microscopía de luz y estereología. La presión arterial (PA) aumentó desde la primera semana de administración de L-NAME. El tratamiento con Losartan, en las dosis de 20 y 40 mg/kg/día, fue eficiente para reducir la PA después de la séptima semana de tratamiento. El remodelamiento cardiaco adverso en el grupo LN se caracterizó por intensa fibrosis intersticial, disminución de la microvascularización miocárdica e hipertrofia y consecuente pérdida de cardiomiocitos. La estructura de la pared de la aorta (densidad por área de núcleos de células musculares lisas y densidad de superficie de lamelas), y la relación media/luz de la arteria mesentérica craneal, también fueron muy alteradas por la administración de L-NAME. Sólo en una dosis igual o mayor que 20 mg/kg/día, el Losartan tuvo efecto benéfico tratando estas alteraciones. En conclusión, tanto el corazón como la pared arterial de ratas deficientes en óxido nítrico, presentan un proceso de remodelamiento acentuado, y éste es eficientemente tratado con Losartan en diferentes dosis. La eficiencia del tratamiento con Losartan en el modelo de bloqueo de la síntesis de óxido nítrico se correlaciona con el efecto hipotensor de la droga, principalmente en las dosis más elevadas.


We studied with morphological tools the effects of different doses of Losartan upon the cardiovascular remodeling in nitric oxide deficient rats. At 15 weeks of age, thirty Wistar rats were separated in six groups: control (C), L-NAME (LN), and four groups were LN was given plus Losartan at different doses (1, 5, 20 and 40 mg/kg/day). The L-NAME was given for 9 weeks, the Losartan administration starting on the 2nd week of experiment. We studied the heart, thoracic aorta and superior mesenteric artery with light microscopy and stereology. The blood pressure (BP) increased since the first week of L-NAME administration, the Losartan treatment at doses of 20 and 40 mg/kg/day was efficient to reduce BP after the 7th week of treatment. The cardiac adverse remodeling in the LN group was characterized by intense interstitial fibrosis, impairment of the myocardial microvascularization, cardiomyocyte hypertrophy and consequent loss of cardiomyocytes. The aortic wall structure (density per area of smooth muscle cell nuclei and surface density of lamellae), and the superior mesenteric artery media/lumen ratio were also strongly affected by L-NAME administration. Only in the dose equal or higher than 20 mg/kg/day Losartan showed beneficial effects treating these alterations. In conclusion, both the heart and the arterial wall of NO deficient rats suffer a marked adverse remodeling process that is efficiently treated by a dose-dependent Losartan administration. The efficiency of Losartan treatment in this model of NO synthesis blockade correlates with the hypotensor effect of the drug mainly in the high dose treatment.
